 The Journal of Biological Chemistryに、村田 美幸副主任の論文「placental growth factor stabilizes VEGF receptor-2 protein in retinal pigment epithelial cells by downregulating glycogen synthase kinase 3 activity」が受理されました。「基礎研究に従事する人間であれば一生に一度は挑戦したい雑誌」とあの控えめな村田先生がおっしゃられて勝負した雑誌です。村田先生、論文アクセプトおめでとうございます!

 VEGFファミリー分子の一つPlGFが、血管新生や血管透過性亢進を惹起する以外に、実は網膜色素上皮細胞(RPE)に対する保護機能も有していることを明らかにした論文です。村田先生は、PlGFをRPEから欠失させるとVEGF-Aの受容体であるVEGFR-2が減少することを発見し、その原因はPlGFを産生できないRPEではVEGFR-2の分解が促進してしまうためであることを突き止められました。そして、その機序にはGSK3という酵素が関わっているという詳細な分子メカニズムも明らかにされました。

 我々眼科医は、抗VEGF製剤の治療効果にばかり注目してしまいがちです。しかし、「抗VEGF製剤が眼組織に対してどのような影響を与えているのか」を知ることは、各疾患に対する同製剤の長期治療成績を考える上で、そして次の創薬を考えていく上でもとても大切なことだと思います。彼女が長年をかけて明らかにしたこの事象は、インパクトファクタ ーのような評価基準だけでは見出せない(JBCもいい雑誌ですが)、科学的探究としての高い価値があると思います。

 そして、良い眼科臨床を志す北大眼科の若い先生たちには、大学院に進んでこの村田先生の指導を受けて、ぜひ「科学的な視点」を養って欲しいと思うのです。

 The paper entitled “Placental growth factor stabilizes VEGF receptor-2 protein in retinal pigment epithelial cells by downregulating glycogen synthase kinase 3 activity”, which vice principal investigator Dr. Murata wrote this year, was accepted by the Journal of Biological Chemistry. We challenged the classic journal because Dr. Murata who is a modest lady told that “This is the journal that one engaging in basic science desires to challenge once in his/her life.” Yes! Big congratulations to Dr. Murata!

 In the brilliant work, Dr. Murata clearly elucidated that PlGF, a mem-ber of the VEGF family proteins, participates in the protection of retinal pigment epithelial cells (RPE) in parallel with induction of neovasculariza-tion and vascular leakage. Dr. Murata found that PlGF ablation reduced the protein level of VEGFR-2 in RPE. She also revealed that PlGF depletion accelerated the degradation of VEGFR-2 protein via downregulating gly-cogen synthase kinase 3 activity in RPE.

 We ophthalmologists are apt to focus on the therapeutic effects of anti-VEGF agents alone. However, I believe it is quite of importance to know both positive and negative impacts of anti-VEGF agents on ocular tissues in order to think over the long-term treatment outcome of the drug and to develop the next-generation therapeutic agents. Although Dr. Murata spent several years to publish her data, the experimental achievement is of much value as scientific exploration, which is never evaluated by popular journal evaluation such as impact factor.

 And, I really would like young doc-tors in our department to go on to postgraduate school and to be educated by Dr. Murata to build their scientific view.
